The vacuum hose comes from the rear intake. It goes to the fuel tap to ensure that the fuel tap only passes fuel when the engine is turning over, creating vacuum.

Bit confusing as you say the pipe (singular) has two other ends?

The vacuum pipe attaches to the petcock assembly at the small side pipe, not the bottom. There are two other pipes that go on the petcock, one for each carburettor fuel inlet. You then have a short overflow pipe and the breather from the tank itself.
Two bikes, still only four cylinders!

Failing that, it's a trip outside with some tools and have a look see and report back for one of us matey.

I'm sure there's a thread with a good explanation but I'm damned if I can find it.

Vacuum pipe should be singular... goes onto the inside of the rear cylinder head doesnt it not? :)

Yep
Rear cylinder diagram 12.3b above shows a view from the left hand side of the bike (when sitting on it) just at the base of the carb inlet.
